Good advice from Franco... let me add:

Don't assign permission manually, create subwebs, assign groups to
subwebs via FP Explorer...
Wouldn't hurt if it was an admin account doing the above...
Add users to groups and add groups to FP Explorer for admin, author,
browse permissions (this is on one of MS
's KB\White papers)...

If something does blow up, like Franco said, it's usually because of
permissions. We've found the safest way (jeez, we feel like hostages)
is to let FP do it all... since then, it's worked like a charm.
